چکیده

Cotton string coated with 3-methylcholanthrene (MCA) was implanted in the uterine cavity of ICR mice to induce endometrial hyperplasia and adenocarcinoma. A low dose (total, 2.5 mg) or a high dose (total, 35 mg) of progesterone was administered to the mice at various times during a period of 4 to 40 weeks after MCA application. After serial labeling with [3H]thymidine, the mice were sacrificed, and thymidine-labeling index values of endometrial hyperpla sia and adenocarcinoma both in the progesterone-treated and the untreated mice were investigated by autoradiographic techniques. In mice that had been implanted with MCA, there was a progressive increase of hyperplasia and neoplasia as a function of time. The low dose of progesterone adminis tered to the mice caused a significant reduction in labeling with [3H]thymidine in nonatypical hyperplasia and moder ate atypical hyperplasia, compared to that for the un treated mice. In marked atypical hyperplasia and adeno carcinoma, irrespective of the histolÃ3gica! grade, labeling was not reduced. With the high dose of progesterone, marked morpholog ical alterations with degenerative changes were observed in atypical hyperplasia and in differentiated adenocarci noma. However, cancer cells were still maximally labeled. The results indicate that the effect of progesterone on nonatypical hyperplasia and moderate atypical hyperpla sia is mitotic arrest, whereas the effect on marked atypical hyperplasia and adenocarcinoma is morphological and cytological alterations.

Background: Endometrial carcinoma (EC) is the most common gynecologic malignancy however, mechanisms underlying its pathogenesis remain obscure. Endometrial carcinoma has been classified into two major categories: type I (related to estrogen or endometrioid adenocarcinoma) and type II (unrelated to estrogen). Estrogen is the main trigger for the abnormal proliferation in the endometrial epithel...

Evaluation of the Effect of Additive Metformin to Progesterone on Patients with Endometrial Hyperplasia

Background and Objectives: Endometrial hyperplasia (EH) is an abnormal overgrowth of endometrium that may lead to endometrial cancer, especially when accompanied by atypia. The treatment of EH is challenging, and previous studies report conflicting results. Metformin (dimethyl biguanide) is an anti-diabetic and insulin sensitizer agent, which is supposed to have antiproliferative and anticancer...

P-120: The Impact of Progesterone and Sildenafil on Endometrial Ultrastructure in Superovulated Mice

Background: Implantation is proceeded by a process referred as adhesion. Adhesion requires maturation of endometrium and blastocyst to confer adherence ability to them. During IVF cycles, endometrial maturation is facilitated by progesterone administrating. Considering vasodilatory effect of Sildenafild, in genital organs, it appears that Sildenafil may have similar effect as progestrone. The a...

Effect of Matrigel on Function and Morphology of Human Endometrial Epithelial Cell in vitro

The importance of extra cellular matrix (ECM) in development and function of different cells has been reported but little is known about its role in human endometrial epithelial cells. The aim of the present study was to examine effects of artificial ECM (Matrigel) and progesterone on the function and morphology of human endometrial epithelial cells in vitro. Methods: Endometrial samples were ...
